There is a post in spn_heavymeta from October 25th of last year, in it ataniell93 brought up the symbol and the connetion to Azazel. She now has a theory on who Ruby represents that I am axiously waiting for.
Angels both fallen and aloft have bee mentioned a few times int he series (many times last season) and there have been one or two references to the Nephilim but other than the Azazel symbol you had to squint to notice them.
Sam I believe (I brought this up early last year I think) is Samael, and Dean was paralleled to Michael in Houses of the Holy. I have some other symbolism stuff in this post: http://deathisyourart.livejournal.com/99925.html
